Determination of the active ingredient in pharmaceutical gel formulation by NIR spectroscopy

Pharmaceuticals of their intended must be thoroughly controlled. The traditional analytical methods are very costly and time consuming. NIR spectroscopy allows to analyze pharmaceutical materials very quickly and with very low costs. First pharmaceutical applications of the NIR spectroscopy appeared with some incuriosity in the late 1960s. Application of NIR in the contemporary pharmaceutical industry is very large. The most common are: the identification of raw materials, identification of active ingredients and excipients in many drug ingredient (tablets, powders, granulates) at the intermediate stage and in the end of the formulation process, the determination of moisture content, hardness, film coating experiences, homogeneity, evaluation of practice size distributions or blending studies.In this work we demostrate suitability of FT-NIR spectroscopy in determination of active ingredient in pharmaceutical gel formulation. The gel spectra were collected with no sample preparation with transflectance module over the spectral range 10000-4000cm-1. Calibration model was built using partial leas square (PLS) regression analysis. Optimum models were found by choosing proper spectral ranges and number of 3 LV factors. Raw spectra were pretreated with Stavitzky-Golay algorithm, and SNV transformation.Results of proposed method shows good predictive ability of the active ingredient in the matrix where pharmaceutical preparation exhibits strong absorbtion in the NIR spectral region.
