    Beta-D-galactosidase (EC 3.2.1.23) catalyze the hydrolysis of terminal non-reducing beta-D-galactose residues in beta-D-galactosides. Cold-active beta-D-galactosidases have recently become a focus of attention of researchers and dairy product manufactures owing to theirs ability to: (I) eliminate of lactose from refrigerated milk for people afflicted with lactose intolerance, (II) convert lactose to glucose and galactose which increase the sweetness of milk and decreases its hydroscopicity, and (III) eliminate lactose from dairy industry pollutants associated with environmental problems. Moreover, in contrast to commercially available mesophilic beta-D-galactosidase from Kluyveromyces lactis the cold-active counterparts could make it possible both to reduce the risk of mesophiles contamination and save energy during the industrial process connected with lactose hydrolysis. A genomic DNA library was constructed from soil bacterium Paracoccus sp. 32d. Through screening of the genomic DNA library on LB agar plates supplemented with X-Gal, a novel gene encoding a cold-active beta-D-galactosidase was isolated. The in silico analysis of the enzyme amino acid sequence revealed that the beta-D-galactosidase Paracoccus sp. 32d is a novel member of Glycoside Hydrolase Family 2. However, owing to the lack of a BGal_small_N domain, the domain characteristic for the LacZ enzymes of the GH2 family, it was decidet to call the enzyme under study BgaL. The bgaL gene was cloned and expressed in Escherichia coli using the pBAD Expression System. The purified recombinant BgaL consists of two identical subunits with a combined molecular weight of about 160 kDa. The BgaL was optimally active at 40 degrees C and pH 7.5. Moreover, BgaL was able to hydrolyze both lactose and o-nitrophenyl-betaD-galactopyranoside at 10 degrees C with Km values of 2.94 and 1.17 mM and kcat values 43.23 and 71.81 s-1, respectively. One U of the recombinant BgaL would thus be capable hydrolyzing about 97% of the lactose in 1 ml of milk in 24 h at 10 degrees C. A novel bgaL gene was isolated from Paracoccus sp. 32d encoded a novel cold-active beta-D-galactosidase. An E. coli expression system has enabled efficient production of soluble form of BgaL Paracoccus sp. 32d.

    W pracy zaprezentowano, nową i szybką metodę analizy komponentów falowodowych. Metoda opiera się na kombinacji metody różnic skończonych, metody dekompozycji dziedziny obliczeniowej, redukcji rzędu modelu oraz dyskretnych rozwinięć funkcyjnych. Przedstawiona technika pozwala na skrócenie czasu analizy układu nawet o dwa rzędy wielkości w stosunku do poprzednich algorytmów.

  • A procedure for the determination of dichloromethane and tetrachloroethene in water using pervaporation and gas chromatography

    In the present study, pervaporation was applied to the determination of tetrachloroethene (PCE) and dichloromethane (DCM) in liquid samples. PCE is the most commonly used solvent in dry-cleaning processes. PCE belongs to group 2A of carcinogens (probably carcinogenic to humans) according to the classification of the International Agency for Research on Cancer (IARC). DCM is also widely used as an industrial solvent for the purification and isolation of intermediates or products. DCM is classified as a "possible" human carcinogen by the IARC. The aim of this study was to evaluate a new procedure for the determination of DCM and PCE in liquid samples based on the pervaporative removal of DCM and PCE from liquid samples as an analyte isolation/enrichment technique, followed by a direct aqueous injection of the extracts onto the column of a gas chromatograph equipped with an electron capture detector (DAI-GC-ECD). The basic parameters of the new PV-DAI-GC-ECD procedure were evaluated.


  • A proposition for integrating elements of game universe by means of behavioral trees

    Modern computer games often involve autonomous beings collaborating and competing with each other to reach their objectives in the game universe. It seems only natural to use agent-oriented approach in such cases. This article identifies the rules of building a game universe as an environment for executing agents. The paper also covers a method for decomposing compound behaviors into behavioral trees. Dividing complex behaviors into smaller and easier to implement ones, each with its own preconditions and finalizing actions, leads to a natural model for defining the way individual agents should act. Because of their structure, behavioral trees are suitable for describing goal-oriented behaviors with conditional branches. This approach leads to a proposition for integrating elements of game universe which is covered in the final part of the paper. Presented solution was implemented in an existing real-time strategy game Nasibu Isle.


  • A prototype of electrochemical sensor for measurement of carbonyl compounds in air

    The paper presents the results of investigation on a prototype sensor for measurement of benzaldehyde and formaldehyde in air. Sensitivity, limit of quantification and coefficient of selectivity of the sensor have been determined as a function of membrane thickness and electrode type using square wave voltammetry (SWV) technique. The working and counter electrodes were made of platinum and gold. Ionic liquids 1-hexyl, 3-methylimidazolium bis (trifluoromethanesulfonyl)imide constituted an internal electrolyte. Polydimethylsiloxane (PDMS) membrane separated gaseous medium from the electrolyte.


  • A research on the optimal conditions for separation and identyfication of the components from the Droseraceae extracts by column liquid chromatography

    The isolation of the components with anticipated biological properties i.e. antimicrobial, fungicidal, antioxidant and perhaps anticancer activity is for many years a one of the biggest issues of modern separation science. Their obtaining based on the isolation of the components with the biological activity from in vitro cultured plants by extraction techniques is often more difficult than their production by the chemical synthesis.The paper presents results of research on the development of the optimal conditions for the separation and identification of components from four carnivorous Droseraceae family plants extracts, such as D. muscipula, D. aliciae, D. binata and D. capensis. The main purpose of the research was to find the most selective chromatographic conditions to separate the naphthoquinones from flavonoids. In the paper the high performance liquid chromatography in the reversed phase (RP-HPLC), was used. The data obtained from separation plant extracts by isocratic elution, gradient elution and step elution was compared in the paper.The paper compares results obtained using gradient elution, isocratic elution and step elution. The use of the isocratic and step elution is favorable in the case of separation of the compounds mixture in the preparative scale chromatography (P-LC) and particularly in the process liquid chromatography. Application of the gradient elution is useful in the study of the plant metabolomics, identification and quantitation of the components of the plants extracts and also quality control and standardization of herbal materials.


  • A robust 18-pulse diode rectifier with coupled reactors

    The article presents the principle of operation and selected results of simulation and laboratory tests of the 18-pulse rectifier system with coupled reactors and small series active power filter. The presented system makes it possible to reduce, especially in distribution supply networks, undesired higher current harmonics. The 18-pulse nature of operation of the rectifier is reached using a set of coupled three-phase network reactors (CDT and CTR). The simultaneous use of the coupled reactors and the small active power filter provides opportunities for reduction of the supply current distortion, especially in case of voltage harmonic distortion and voltage unbalance.


  • A selection of the optimal eluent components for separation of carnivorous plants metabolites by planar chromatography chromatography

    Abstract: The development of the new procedures of the separation components with the complicated chemical structure from natural complex matrix by column chromatography is very difficult and in many cases expensive. Therefore to pre-select the conditions of the separation process the use of thin layer chromatography (TLC) is proffered. The possibility of proceeding the chromatographic process for several mixtures on single plate, significantly reduces time and amount of the reagents needed for completing the "set" of the separations in the tested chromatographic conditions. Another advantage is the simplicity of using a so-called orthogonal separation - a separation of few mixtures in the same TLC plate in two perpendicular directions with use of two different eluents. This allows doubling the retention data of separated components achieved for analyzed stationary phase. The use of suitably chromatographic system in connection with the specific biological and chemical methods of detection, allows getting preliminary information about presence and retentions parameters of biologically active components occurring in separated mixtures.The paper presents an analysis of the retention data obtained for reversed phase TLC with two-, three-component component eluent for selection of the optimal selectivity of the chromatographic system. In the paper a results of comparison of the sorption properties of different stationary phases are also provided. An influence of the acid addition to the eluent on the so-called peak "tailing" of the flavonoids is also discussed

W artykule zaproponowano prosty mechanizm umożliwiający rozstrzyganie takich konfliktów w drodze automatycznie realizowanych negocjacji, którego wdrożenie pozwoli znacznie rozszerzyć zakres stosowania standardu ebXML.


  • A solution of non-linear differential problem with application to selected geotechnical problems

  • A spice equivalent circuit for modeling the performance of dual frequency echo-sounder

    The paper presents novel network equivalent circuit of piezoceramic circular disc transducers that takes into account thickness and radial mode of vibrations. The starting point of the analysis is 4-port description of circular disc element representing the solution of wave equation set in radial and thickness directions. The approximate solution for harmonic case is represented in the form of 4x4 matrix, which is synthesised and implemented in circuit analysis software. The network is extended with acoustical port that allows for pressure calculations in the similar manner as when measuring echo-sounder transmitting and receiving parameters. the model is especially useful for simulating complete electronic circuit dual frequency echo-sounder based on single circular disc transducer.

    W pracy przedstawiono wyniki wstępnych badań ukierunkowanych na wykrycie podstawowych fermonów wydzielanych w sytuacji zagrożenia (fermony alarmowe) przez różne organizmy żywe.W badaniach zastosowano takie techniki jak TD-GC-MS, SPME-GC-MS stwierdzono występowanie fermonów należących do takich klas związków organicznych jak alkohole, węglowodory nasycone, aldehydy, kwasy karboksylowe i ketony.

    W artykule przedstawiono przebieg międzynarodowej konferencji "20th IEEE International Symposium on Industrial Electronics", zorganizowanej przez Wydział Elektrotechniki i Automatyki PG pod patronatem IEEE Industrial Electronics Society (IES) w dniach 27-30.06.2011 r. W konferencji uczestniczyło ok. 380 uczestników, w tym ponad 80% z zagranicy. W artykule sprawozdano wygłoszone seminaria dydaktyczne, referaty plenarne i najlepsze nagrodzone referaty. Przedstawiono obchody 60-lecia IES z udziałem byłych i obecnych prezydentów stowarzyszenia.

    I provide a guide to Gdańsk (Danzig) and some of its suburbs, focusing on sites of particular interest to physicists. These include the Town Hall with the old Gdańsk standards of length at its entrance, the solar dial of 1588 on its corner, and its bell tower; the Naturalists Society; the medieval harbor crane; the medieval astronomical clock in St. Mary's Church; the late nineteenth-century lighthouse and time ball; and sites associated with Nicolaus Copernicus (1473-1543), Johannes Hevelius (1611-1687), Daniel Gabriel Fahrenheit (1686-1736), and Daniel Gralath (1708-1767). I also comment on the history of and physical research being carried out today at the Technical University of Gdańsk and the University of Gdańsk.
